摘要
目的检测子宫内膜样腺癌组织中真核起始因子-4A(eIF-4A)、细胞周期蛋白D1(Cyclin D1)的表达,探讨其临床意义。方法采用免疫组织化学方法检测120例子宫内膜样腺癌组织及40例正常子宫内膜组织中eIF-4A及Cyclin D1蛋白的表达,并分析两者之间的相关性。结果子宫内膜样腺癌组织eIF-4A和Cyclin D1蛋白的阳性表达率明显高于正常子宫内膜组织(χ2=38.72、46.73,P〈0.01)。eIF-4A蛋白在子宫内膜样腺癌组织中表达与肿瘤临床病理分期、淋巴结转移、组织分化程度有关(χ2=4.36-13.36,P〈0.05);Cyclin D1蛋白在子宫内膜样腺癌组织中表达与肿瘤组织分化程度及淋巴结转移有关(χ2=24.73、5.24,P〈0.05)。子宫内膜样腺癌组织eIF-4A与Cyclin D1的表达呈显著正相关(r=0.824,P〈0.01)。结论 eIF-4A及Cyclin D1可能参与了子宫内膜样腺癌发生、进展及淋巴转移的过程,在此过程中eIF4A对Cyclin D1表达可能起调控作用,两者有望成为预测子宫内膜样腺癌恶性程度的重要生物学指标。
Objective To detect the expressions of eukaryotic initiation factor-4A(eIF-4A)and Cyclin D1 in endometrioid adenocarcinoma(EA)and explore their clinical significance. Methods Immunohistochemistry was used to detect the expressions of eIF-4Aand Cyclin D1 in 120cases of EA and 40 cases of normal endometrium.The correlation between the two expressions was analyzed. Results The positive expression rates of eIF-4Aand Cyclin D1 in EA tissue were higher than that in normal endometrium(χ2=38.72,46.73;P〈0.01).The expression of eIF-4Ain EA was related to the clinical stage,lymph node metastasis and histological differentiation(χ2=4.36-13.36,P〈0.05),and that of Cyclin D1 in EA was associated with histological differentiation and lymph node metastasis(χ2=24.73,5.24;P〈0.05).There was a positive correlation between Cyclin D1 and eIF-4Ain EA(r=0.824,P〈0.01). Conclusion eIF-4Aand Cyclin D1 are likely involved in the process of carcinogenesis,development and lymphatic metastasis of endometrioid adenocarcinoma.In the process of occurrence and progression of this malignancy,the eIF-4Amay regulate the expression of Cyclin D1,these two expressions are expected to become significant biological parameters for predicting the degree of malignancy of endometrioid adenocarcinoma.
