膀胱癌间质蛋白质表达谱及转移潜能评价分析

摘要 目的使用放射性核素相对标记与绝对定量技术(iTRAQ法)分析不同转移潜能浸润性膀胱癌间质蛋白的表达谱特征,并与尿液中蛋白质表达谱进行对比,探讨从尿液中建立肿瘤生物标记组用于评价膀胱癌转移潜能的可能性。方法首先应用激光捕获显微切割技术(LCM),获取纯化的高、中、低危组肿瘤细胞标本,然后根据iTRAQ法分别鉴定高、中、低转移风险浸润性膀胱癌细胞蛋白质表达谱,并分析其差异表达。结果 3组肿瘤细胞标本中有943个蛋白共同表达,根据基因本体论进一步分析表明,943个蛋白质中具有生物学途径、细胞成分注解的蛋白质分别为760、804个。通过与国际蛋白质索引的完整列表进行对比,生物学途径中浓集-缺失的分支术语为55/11,细胞成分中浓集-缺失的分支术语为43/8。结论 LCM联合iTRAQ法分析不同转移潜能浸润性膀胱癌间质蛋白的表达谱特征,结果可靠、有效;细胞的生物学途径和亚细胞结构决定膀胱转移潜能。 Objective By using isobaric tags for relative and absolute quantitative(ITRAQ)technique,the features of protein expression profile of different metastatic potential in invasive bladder cancer were analyzed and compared with the expression profile of protein in urine to explore the possibility of creating a tumor biomarker from urine for assessing the metastatic potential of bladder cancer. Methods Laser capture microdissection(LCM)technique was used first to obtain purified tumor cell samples from high-,medium-and low-risk groups,which were then identified for the protein expression profile in the three groups of bladder cancer with different metastatic and invasive risks-based on ITRAQ technique-and the differences of expressions were analyzed. Results Of tumor cell specimens in the three groups,there were 943 protein co-expressions.A further analysis-based on gene ontology(GO)-indicated the proteins that with biological pathways or cellular component annotation were 760 and 804,respectively.Compared with the complete list of International Protein Index(IPI),the branch terms of concentration-deletion in biological pathways were 55/11,and that in components of cells were 43/8. Conclusion The result of protein expression profile is reliable and valid obtained by using combined LCM with ITRAQ analysis.Biological pathways of the cells and subcellular structures determine the metastatic potential of the bladder cancer.
