介入化疗前后宫颈癌组织pS53及pS642表达及意义 被引量:1

EXPRESSIONS AND SIGNIFICANCE OF PS53 AND PS642 IN CERVICAL CARCINOMA TISSUE BEFORE AND AFTER INTERVENTIONAL CHEMOTHERAPY

摘要 目的探讨细胞有丝分裂抑制酶Wee1第53及642位点丝氨酸磷酸化蛋白(pS53、pS642)在动脉栓塞化疗前后宫颈癌组织中表达的变化及意义。方法采用免疫组织化学方法检测50例ⅠB-ⅡB期宫颈癌动脉栓塞化疗前后组织标本和40例正常宫颈组织标本中pS53及pS642的表达,并分析两种蛋白表达的变化与临床治疗效果的关系。结果正常宫颈组织中pS53及pS642的表达低于动脉栓塞化疗前宫颈癌组织,差异有显著性(χ2=42.50、28.10,P〈0.01)。动脉栓塞化疗后宫颈癌组织中pS53及pS642的表达较动脉栓塞化疗前明显下降,差异有显著性(χ2=17.36、19.84,P〈0.01)。动脉栓塞化疗前后宫颈癌组织中pS53及pS642的表达变化与化疗效果呈负相关(r=-0.328、-0.456,P〈0.01)。结论宫颈癌组织中pS53及pS642的异常激活及高表达与宫颈癌的发生发展密切相关。动脉栓塞化疗后宫颈癌组织中pS53及pS642的失活及低表达与其效果呈明显负相关,故两者可成为临床治疗效果判断的重要指标。 Objective To investigate the expressions and significance of phosphorylation of serine 53(pS53)and 642 of the cell mitotic inhibitor kinase Wee1(pS642)in cervical carcinoma tissue before and after arterial chemoembolization(ACE).Methods By employing immunohistochemical method,samples from 50 cases of stageⅠB-ⅡB cervical cancer were detected for expressions of pS53 and pS642-before and after ACE-and 40 cases of normal tissue of cervix served as controls.The correlation between the treatment effectiveness and the expressions of the two proteins was analyzed. Results In normal cervical tissue,the expressions of pS53 and pS642were lower than that in the cancer tissue before ACE(χ2=42.50,28.10;P〈0.01).After ACE,the expressions of pS53 and pS642decreased(χ2=17.36,19.84;P〈0.01).The expressions of pS53 and pS642-before and after ACE-were negatively correlated with the effect of chemotherapy(r=-0.328,-0.456;P〈0.01). Conclusion The abnormal activation and high expressions of pS53 and pS642in cervical carcinoma tissue are closely related to the occurrence and development of the cancer.The deactivation and lower expression of pS53 and pS642after arterial chemoembolization in cervical carcinoma tissue were negatively correlated with the efficacy of chemotherapy,so,the two expressions can be used as important parameters for predicting the outcomes of therapy.
