肝癌组织中脯氨酸羟化酶3(PHD3)的差异表达对预测原发性肝癌切除术后预后的价值 被引量:3

摘要 目的检测脯氨酸羟化酶3(prolyl hydroxylase 3,PHD3)在肝癌及癌旁组织中的表达情况,探讨其对原发性肝癌切除术后患者预后的预测价值。方法收集本院2006年进行肝切除术后病理确诊为肝细胞肝癌的标本325例,制作成组织芯片。采用免疫组化法检测肝癌及癌旁肝组织中PHD3的表达水平,并比较PHD3表达水平与肝癌患者无瘤生存时间及总体生存时间的相关性。应用Kaplan-Meier法、χ2检验及t检验分析PHD3的预测价值及与临床病理参数的关系。在325例患者组织标本中随机选取无复发转移和复发转移患者各6例的肝癌组织标本,提取组织蛋白进行Western blot检测。结果免疫组化显示,肝癌细胞中PHD3主要在胞质表达,核内也有表达。癌组织中PHD3的表达水平与与肝癌患者的生存预后显著正相关,与患者性别、微血管侵犯、肿瘤大小和甲胎蛋白不相关。癌旁组织中PHD3的表达与预后无显著相关。Western blot检测发现6例无复发患者的PHD3蛋白表达明显高于复发患者。结论肝癌组织中PHD3的表达可以有效预测原发性肝癌切除术后患者的预后。 Objective To study the prognostic value of the intratumoral and peritumoral expression of prolyl hydroxylase 3 (PHD3) in hepatocellular carcinoma (HCC) after curative resection.Methods A total of 325 cases liver resection specimens diagnosed as HCC in the year of 2006 were made into a tissue microarray.Immunohistochemistry was used to detect the expression of PHD3 in HCC and adjacent normal liver tissues of this tissue microarray.Relationship between PHD3 expression level and disease-free survival and overall survival in HCC patients was compared.The prognostic value of PHD3 and its realationship with other clinicopathological factors were analyzed by the application of KaplanMeier method,x2 test and t test.Western blot analysis was used to detect PHD3 expression in 12 cases of HCC tissues including 6 non-recurrent cases and 6 recurrent cases which were randomizedly selected from 325 specimens.Results Immunohistochemical results showed that intratumoral PHD3 expression level was significantly correlated with the prognosis of HCC patients.And it was not associated with patients' gender,microvascular invasion,tumor size,and alpha-fetal protein.No significant correlation was found between peritumoral PHD3 expression and HCC patient prognosis.Western blot results showed that intratumoral PHD3 expression of recurrent cases was higher than that of non-recurrent cases.Conclusions Intratumoral PHD3 expression can effectively predict the prognosis of HCC patients after curative resection.
