摘要
目的研究不同分化程度肾癌中MMP-26的表达,并探讨其与肾癌分化程度的相关性。方法通过免疫组织化学(ABC法)检测60例不同分化程度肾癌和20例肾脏良性病变组织中MMP-26的表达情况,并采用RT-PCR技术检测20例新鲜肾癌组织和10例肾脏良性病变组织MMP-26的mRNA表达。结果 MMP-26在肾癌组织的阳性表达率为80%,在肾脏良性病变组织为50%(P<0.05);G1、G2、G3级肾癌组织中MMP-26的阳性表达率分别为88.5%、89.5%和53.3%,G3级显著低于G1和G2级(P<0.05),但G1、G2级之间的差异无统计学意义(P>0.05)。肾癌组MMP-26 mRNA的表达为(0.83±0.126),而肾脏良性病变组MMP-26 mRNA的表达为(0.60±0.035)(P<0.05);G1、G2、G3肾癌组织中MMP-26 mRNA的表达分别为(1.13±0.104)、(0.90±0.072)和(0.80±0.031),组间两两比较,差异有统计学意义(P<0.05)。结论 MMP-26在肾癌组织中呈高表达,并与肾癌的分化程度负相关,有望作为肾癌病情评估和预后预测的重要参考指标。
Objective To investigate the expression of matrix metalloproteinases-26 (MMP-26) in renal carcinoma tissue, and to probe its relationship with the differentiated degrees of renal carcinoma tissue. Methods Immunohistochemistry method (ABC) was used to detect the MMP-26 expression of 60 cases of renal carcinoma tissues in different differentiated degrees and 20 cases of benign lesion tissues. Reverse transcription polymerase chain reaction (RT-PCR) was applied to detect the mRNA expression of 20 cases of surgical fresh tissues with renal carcinoma and 10 cases of benign lesion tissues. Results The positive expression rate of MMP-26 was 80%in renal carcinoma pathological tissues, but was 50%in benign renal pathological tissues (P〈0.05). The positive expression rate of MMP-26 was 88.5%, 89.5%and 53.3%respectively in G1, G2 and G3 differentiation degree;It was statistically lower in G3 than in G1 and G2 (P〈0.05), but its difference between G1 and G2 was of no significance (P〉0.05). The mRNA expression of MMP-26 was (0.83±0.126) in renal carcinoma tissues, and (0.60±0.035) in benign le-sion tissues (P〈0.05). The mRNA expression of MMP-26 was respectively (1.13±0.104), (0.90±0.072) and (0.80±0.031) in G1, G2 and G3 of renal carcinoma tissues;Difference between each two groups had statistical significance (P〈0.05). Conclusion MMP-26 was highly expressed in renal carcinoma, and negatively correlated with the differentiation degrees of renal carcinoma.It would serve as an important indicator for evaluating severity and prognosis of renal carcinoma.
