BMP-2对人卵巢浆液性囊腺癌HO8910细胞浸润能力的影响及机制研究

摘要 目的检测骨形态发生蛋白-2(BMP-2)对人卵巢浆液性囊腺癌HO8910细胞的浸润能力有无影响,并探讨其可能的影响机制。方法 2008至2009年于中国医科大学盛京医院应用细胞黏附实验、cross-river实验和matrigeltranswell实验观察BMP-2对HO8910细胞浸润能力的影响,并检测BMP-2作用后HO8910细胞中MMP-2、TIMP-2的变化。结果 BMP-2作用后HO8910细胞黏附能力增强,cross-river时间延长,穿过matrigel-transwell侵袭模型的细胞数量减少,且MMP-2表达较前减弱,TIMP-2表达较前增强。结论 BMP-2抑制HO8910卵巢癌细胞浸润能力。 Objective To verify the influence of bone morphogenetic protein-2 (BMP-2) on the invasion capacity of human ovarian serous cystadenoma cell HO8910, and to determine the possible mechanism.Methods Cell adhesion experiment, cross-river experiment and matrigel-transwell assay were performed to observe the influence of BMP-2 on the invasion ability of HO8910 cells. The changes of MMP-2 and TIMP-2 in HO8910 cells following BMP-2 in- tervention were also detected in Sheng Jing Hospital of China Medical University between 2008 and 2009. Results Following the BMP-2 intervention, the adhesion capacity of HO8910 cells was shown to increase, the cross-river time was longer and the cells that could cross matrigel-transwell model were decreasing. Moreover, MMP-2 expression was attenuated while TIMP-2 expression was enhanced.Conclusion BMP-2 can inhibit the inva- sion capacity of HO8910 cells.
