苦瓜冻干超微粉调节肥胖大鼠胰岛素抵抗及其作用机制研究 被引量:7

Mechanism Research of Bitter Melon Lyophilized Superfine Grinding Powder on Insulin Resistance in Nutritional Obese Rats

摘要 目的:研究苦瓜冻干超微粉(BLSP)对营养型肥胖大鼠胰岛素抵抗的调节作用及其机制。方法:通过饲喂高脂饲料的方法建立营养型肥胖模型大鼠,分别灌胃BLSP[200,400 mg/(kg bw)]、吡格列酮[10 mg/(kg bw)]和水[4 mL/(kg bw)]4周后,测定其糖耐量、血清胰岛素、血脂水平等指标。结果:与肥胖模型组相比,BLSP能显著减缓肥胖大鼠体重的增速,增加其糖耐量,降低血清胰岛素、甘油三酯和总胆固醇含量,减缓肝脏脂变,其效果优于吡格列酮。结论:BLSP具有改善营养型肥胖大鼠的胰岛素抵抗症状和保护肝脏的作用。其作用机制可能与促进外周组织吸收利用葡萄糖,减弱脂肪细胞甘油三酯的水解和增加肝脏LPL和HL活性有关。 Objective: Study the effect of bitter melon lyophilized superfine grinding powder(BLSP) on insulin resistance in high-fat diet(HFD)-induced obese rats. Methods: We induced the obese rat model by feeding high fat diet,then treated these rats with BLSP [200, 400 mg/(kg bw)], pioglitazone [10 mg/(kg bw)] and water [4 mL/(kg bw)] respectively for 4 weeks, and finally determinated the levels of glucose tolerance, serum insulin, serum lipid profiles and others. Results: BLSP showed a higher anti-obesity activity with lower rate of increase of obese rats' body weights, reversing HFD-induced glucose intolerance, ameliorating the hyperinsulinemia, decreaseing the levels of TG and CHOL,and inhibiting the steatosis of liver compared with those in the nontreated HFD group. Conclusion: BLSP can ameliorate the symptoms of insulin resistance in obese rats and protect the liver, which will provide a reference for direct utilization of bitter melon as a suitable health food to relieve symptoms of obesity. The mechanism may be related with BLSP can stimulate glucose uptake and utilization by peripheral tissues, reduce the triglyceride hydrolysis of adipocytes and increase the activity of LPL and HL in liver.
