双黄连粉针剂对多重耐药大肠埃希菌菌体内药物蓄积水平的影响 被引量:2

Influence of Shuanghuanglian Injectable Powder on drug accumulation level in body of multi-drug-resistant Escherichia coli

摘要 目的观察双黄连粉针剂对多重耐药大肠埃希菌菌体内药物蓄积水平及其细菌结构的影响。方法采用荧光分光光度法观察双黄连粉针剂作用后柔红霉素在多重耐药大肠埃希菌体内聚集水平的改变;透射电镜观察双黄连粉针剂作用后多重耐药大肠埃希菌细菌结构的改变。结果亚抑菌浓度双黄连分别预处理4,6,8 h后,大肠埃希菌菌体内柔红霉素蓄积程度较空白对照增强(P〈0.05),其中6-8 h时较为明显;1/2,1/4,1/8最低抑菌浓度(MIC)的双黄连预处理6h后,大肠埃希菌细菌内柔红霉素蓄积程度较空白对照增强(P〈0.05),且不同浓度双黄连预处理细菌后柔红霉素蓄积程度改变具有差异(P〈0.05)。透射电镜观察,双黄连作用后,多重耐药大肠埃希菌出现分裂相减少、胞膜缺损、胞质流失等改变。结论双黄连粉针剂可以增加柔红霉素在细菌内聚集,呈现量效和时效关系,并且能够破坏大肠埃希菌细胞膜结构的完整性。 Objective To observe the influences of Shuanghuanglian Injectable Powder on drug accumulation level in the body of multi- drug- resistant Escherichia coli(E.coli) and E.coli structure.Methods The changes of daunorubicin accumulation level in the body of multi- drug- resistant E.coli were observed by using fluorospectrophotometry after affected by Shuanghuanglian Injectable Powder.The changes of E.coli structure were observed by using transmission electron microscope(TEM).Results After pretreatment with sub- MIC Shuanghuanglian Injectable Powder for 4 h,6 h and 8 h respectively,daunorubicin accumulation level was increased in the body of E.coli than that in blank control group(P〈0.05),which was more significant after 6 h-8 h.After pretreatment with 1/2 MIC,1/4 MIC or 1/8 MIC Shuanghuanglian Injectable Powder for 6 h,daunorubicin accumulation level was increased in the body of E.coli than that in blank control group(P〈0.05),and there was difference among different doses of Shuanghuanglian Injectable Powder(P〈0.05).The observation of TEM showed decrease of mitotic phase,defect of cell membrane and loss of cytoplasm.Conclusion Shuanghuanglian Injectable Powder can improve daunorubicin accumulation in the body of E.coli and destroy the integrity of cell membrane of E.Coli in dose-efficacy and time-efficacy manners.
