摘要
为研究番茄ARF2基因表达模式及其调控番茄器官发育的分子机理,利用PCR技术扩增了SlARF2转录起始点上游2462bp的片段,利用农杆菌转化获得SlARF2∶∶GUS融合基因的番茄转基因植株。启动子序列分析表明,该启动子片段含有MeJA、ETH、GA和SA等激素相应的顺式作用元件。GUS染色结果表明,ARF2在生长点、幼叶基部、主茎的表皮毛中表达强烈。暗示SlARF2可能参与调控这些器官的发育过程。
For the study on the expression profile of SlARF2 and its regulatory mechanism in tomato organ development, promoter analysis of SlARF2 was used. A 2462bp fragment upstream of transcription initiation was amplified from tomato genomic DNA by PCR. This fragment was employed to drive GUS reporter genes, and was introduced into tomato by using Agrobacterium-mediated transformation. Sequence contained several putative cis-elements responding to phytohormones (MeJA,ETH,GA and SA) predicted with online promoter analysis software. The GUS results indicated ARF2 was expressed intensely in meristem, basal part of young leaves and epidermal hairs of stem. This result revealed that SlARF2 may participate in regulating the development of these organs.
