摘要
研究不同培养条件对拮抗酵母菌0732-1产生抗菌物质的影响,明确0732-1产生抗菌物质的最适培养条件。以哈密瓜细菌性果斑病菌为指示菌,采用抑菌圈法测定不同培养条件下0732-1发酵液的抑菌活性。结果表明:酵母菌0732-1产生抗细菌物质(ABS)最适碳源为果糖,其次是葡萄糖;以葡萄糖为碳源时,最适氮源为硫酸铵。当酵母菌0732-1接种入PSB培养液中,培养条件为初始pH为9.0;装样量为80 mL(250 mL三角瓶);接种量为10%;培养温度20℃;培养96 h时,发酵液中ABS的抑细菌活性最强,且随着发酵液随发酵液pH值的降低而抑细菌活性增强。
The effects of medium components and other culture conditions on the growth and antibacterial substances production of the yeast strain 0732-1 were investigated to determine the optimum culture conditions.Acidovorax avenae subsp.citrulli(Aac) was used as indicator bacteria,and the method of inhibition zone was used for determining the antibacterial activity of 0732-1 fermentation liquid under different culturing conditions.The results showed that the yeast strain 0732-1 could use six kinds of nitrogen sources and eight kinds of carbon sources for growth, and could utilize each of fructose,glucose, sucrose,maltose, ethanol and glycerin for ABS production.The optimum carbon source was fructose,followed by glucose.When glucose was chosen as carbon source,ammonium sulfate was the optimum nitrogen source for ABS production.The optimum culture factors affecting ABS production by the yeast strain 0732-1 were the amount of PSB being 80 mL in 250 mL flasks;the initial pH value was 9.0;the inoculum was 10%(V/V);the temperature was 20 ℃ and the culture time was 96 h,and then the antibacterial activity enhanced as pH value of the fermentation liquid low.
