摘要
目的研究糖基化终末产物(AGEs)对真皮成纤维细胞(Fb)细胞核因子κB(NF-κB)活化的影响及普罗布考的保护作用。方法体外分离培养Fb,分为对照(Con)组、AGE-BSA组及普罗布考干预(Probucol)组。水溶性四唑盐(WST)法检测细胞增殖,免疫荧光染色检测NF-κB的活化、实时定量PCR检测MCP-1mRNA表达。结果与Con组相比,AGE-BSA组细胞增殖能力减退,NF-κB表达增加,细胞MCP-1mRNA表达升高(P<0.05或P<0.01);与AGE-BSA组相比,Probucol组细胞增殖能力增强,NF-κB表达下降,细胞MCP-1mRNA表达下调(P<0.05或P<0.01)。结论普罗布考对AGE-BSA诱导的Fb的保护作用可能是通过促进Fb增殖,抑制NF-κB活化及减低MCP-1表达来实现的。
Objective To investigate the influence of advanced glycation end products(AGEs) on the activation of NF-κB in dermal fibroblasts and the probucol protection against AGEs.Methods Fibroblasts were isolated and cultured in vitro,and then divided into three groups:control group,AGE-BSA group,probucol group.The proliferation of fibroblasts was determined by WST and the activation of NF-κB was observed by immunofluorescence microscope.The mRNA expression of MCP-1 was detected by Real-time PCR.Results As compared with control group,AGE-BSA group showed that the proliferation of fibroblasts was decreased in AGE-BSA group,but the expression of NF-κB and the mRNA expression of MCP-1 were increased (P<0.05 or P<0.01) ;As compared with AGE-BSA group,probucol showed that the proliferation of fibroblasts was increased,while the expression of NF-κB and the mRNA expression of MCP-1 were reduced(P<0.05 or P<0.01).Conclusion Protective effect of probucol aganist proliferation of fibroblasts induced by AGEs may be related with its effect on inhibition of the activation of NF-κB,promotion of the proliferation of fibroblasts and reduced expression of MCP-1.
