正交实验优化“神农”香菊顶芽的离体培养条件

Optimization of Culture Conditions in vitro Dendranthema indicum var.aromaticumBuds of Orthogonal Test

以湖北神农架地区采集的"神农"香菊顶芽为外植体,研究了不同消毒剂种类及方法的灭菌效果,通过芽增殖培养基和生根培养基的激素配比试验,筛选出"神农"香菊试管苗快速繁殖的最佳培养基组成。结果表明:"神农"香菊顶芽的最佳消毒方案为75%Alcohol 30s+0.1%HgCl22min+2%NaClO38min;顶芽最适增殖培养基为MS+0.7mg/L 6-BA+0.5mg/L KT+0.3mg/L NAA,生根的最适培养基为1/2MS+0.5mg/L NAA。

Taking buds of Dendranthema indicum var. aromaticum introduced from Shennongjia of Hubei as the explants, the sterilizing effect was studied through different disinfectors and methods, by hormone combination test on sprout medium and rooting medium, rooting hormone ratio were studied to selected the best medium for rapid propagation of Dendranthema indicum var. aromaticum were studied. The results showed that the suitable disinfectant was 75 % Alcohol 30 s＋0. 1% HgCl2 2 min＋2% NaClO3 8 min; the suitable sprout medium was MS＋0. 7 mg/L 6-BA＋0. 5 mg/L KT＋ 0. 3 mg/L NAA;the suitable rooting medium was 1/2 MS＋0. 5 mg/L NAA.
