增殖抑制基因和血管内皮生长因子在乳腺癌组织中的表达及与微血管密度的关系 被引量:4

摘要 目的探讨新的增殖抑制基因(HSG)以及血管内皮生长因子(VEGF)在乳腺癌中的表达及与微血管密度(MVD)的关系。方法应用免疫组织化学方法检测唐山市工人医院60例乳腺癌标本、20例乳腺纤维腺瘤及20例正常乳腺组织标本中HSG和VEGF的表达,并用CD34标记乳腺癌组织中的微血管,采用图像分析进行微血管密度(MVD)测定。结果 HSG在乳腺纤维腺瘤和正常乳腺组织中高表达,而在乳腺癌组织中低表达,差异有显著性(P<0.05);乳腺癌组织中,MVD在HSG阳性表达组低于阴性表达组,差异有显著性(P<0.05);VEGF在乳腺纤维腺瘤和正常乳腺组织中低表达,在乳腺癌组织中高表达,且Spearman等级相关分析显示,在乳腺癌组织中,HSG与VEGF表达呈负相关(r=-0.795,P<0.05);MVD在VEGF阳性表达组高于阴性表达组,差异有显著性(P<0.05);另外,在乳腺癌组织中MVD的表达与肿瘤大小和淋巴结转移有关(P<0.05)。结论 HSG在某种程度上可以抑制肿瘤微血管的生成而降低乳腺癌的转移。可见联合检测HSG和VEGF与MVD的表达有利于更好地判断乳腺癌的进展及预后。 [ Objective ] To investigate the expressions of the novel hyperplasia suppressor gene (HSG) and vascu- lar Endothelial Growth Factor (VEGF) in breast cancer and their correlations with tumor micro-vessel density (MVD). [ Methods ] Immunohistochemistry method was used to detect the expressions of HSG and VEGF in 60 cases of breast cancer samples, 20 cases of breast fibro-adenoma samples and 20 cases of normal breast tissues samples. And MVD was detected by CD34 antibody and MVD measurement was based on Image Analysis. [ Results ] The ex- pressions of HSG and MVD were higher in breast fibro-adenoma and normal breast tissues. The expressions of HSG were lower in breast cancer tissues. The difference was statistically significant (P 〈0.05). The expressions of MVD were lower in the positive expression of HSG group than the negative expression of HSG group. The difference was statistically significant (P 〈0,05). The expressions of VEGF were lower in breast fibro-adenoma and normal breast tissues. The expressions of VEGF were higher in breast cancer tissues. Spearman rank correlation analysis showed that the expressions of HSG was negatively correlated with VEGF(r =-0.795, P 〈0.05). The expressions of MVD were higher in the positive expression of VEGF group than the negative expression of VEGF group. The difference was statistically significant (P 〈0.05). And the expressions of MVD were associated with the tumor size and lymph node metastasis (P 〈0.05). [ Conclusions ] In the way, HSG can suppress tumor micro-vessel growth and reduce the rate of breast cancer metastasis. Combined detection of HSG, VEGF and MVD was conducive to better evaluate develop- ment and prognosis of breast cancer.
