摘要
[目的]建立液相色谱对食品中的4种黄曲霉毒素B1、B2、G1、G2进行定性和定量分析的检测方法.[方法]试样经过甲醇+水提取,滤液经过含有黄曲霉毒素特异抗体的免疫亲和层析柱净化,以甲醇通过免疫亲和层析柱洗脱,通过带荧光检测器的高效液相色谱仪柱后碘溶液衍生测定黄曲霉毒素的含量.[结果]试验表明,4种黄曲霉毒素在3.0 ~30.0 ng/ml和10.0 ~50.0 ng/ml质量浓度范围内线性关系良好,相关系数均不小于0.999;4种黄曲霉毒素的检出限均为0.25 μg/kg.在3个不同的加标水平下,4种黄曲霉毒素的回收率在70% ~ 99%,相对标准偏差为分别为1.7% ~7.6%.[结论]该方法能有效检测出多种食品中4种黄曲霉毒素的残留量,且稳定性好,结果可靠.
[Objective] A liquid chromatography method was established for the simultaneously qualitative and quantitative analysis of aflatoxins B1,B2,G1 and G2.[Method] Samples were sequentially extracted by methanol-water,cleaned up with immunoaffinity chromatography column,tested by fluorescence detection and post column derivatization of iodine solution.[Result] The results showed that the method showed good linear relationships in the range of 3.0-30.0 ng/ml and 10.0-50.0 ng/mlL,with correlation coefficients no less than 0.999.Their quantitation limits were 0.25 μg/kg.The mean recoveries ranged from 70% to 99% with relative standard deviations of 1.7%-7.6%.[Conclusion] The proposed method was sensitive and reliable,and was successfully applied in the determination in food.
