黄河三角洲植物真菌的分离、活性菌株筛选及活性产物鉴定 被引量:2

Isolation and antibiotic screening of fungi associated with plants from Yellow River Delta and identification of the active metabolites

摘要 目的从黄河三角洲植物中分离真菌,筛选具有抗菌或抗肿瘤活性菌株,分离鉴定活性成分。方法蔗糖密度梯度法分离菌株,对其发酵物进行抗菌活性和细胞毒活性筛选,采用硅胶柱色谱、凝胶柱色谱对发酵产物进行分离、纯化,运用核磁共振、质谱等手段鉴定化合物的结构。结果从黄河三角洲的9种植物样品中分离纯化真菌136株,筛选得到具有抑菌活性菌株25株、具有细胞毒活性菌株17株,并从1株枝孢属真菌Cladosporiumsp.OUCMDZ-2046的发酵产物中分离鉴定了1个对白色念珠菌和人乳腺癌细胞株MCF-7具有抑制作用的化合物:桔青霉素。结论黄河三角洲的植物真菌具有开发为药用活性菌株的潜力。 Objective To isolate fungi from the Yellow River Delta plants, to screen antibiotic fungal strains, and to identify the active microbial products. Methods The isolation of fungi was performed by sucrose density gradient, the antitumor activities and antibacterial activities of the microbial products were assayed by MTT and 2-fold dilution method, respectively, and the active compounds were purified and identified by column chromatography over silica gel and Sephadex LH-20, and analysis of NMR and ESI-MS, respectively. Results 136 fungal strains were isolated from Yellow River Delta plants, 25 of them showed antimicrobial activities against Candida albicans and Staphylococcus aureus, and 17 of them showed cytotoxic activities against MCF-7 cells. Citrinin was identified from the fermentation broth of Cladosporium sp. OUCMDZ-2046. And eitrinin showed the growth inhibitions of C. albicans and MCF-7 breast cancer cells. Conclusion Plant fungi from Yellow River Delta are potential resources for drug discovery and are worthy of being further studied.
