摘要
目的 研究西沙海绵Agelas aff.nemoechinata的化学成分。方法 利用Sephadex LH-20凝胶色谱、硅胶色谱、ODS色谱、HPLC等手段对化学成分进行了分离纯化;通过理化性质,波谱分析方法结合文献对照,鉴定化合物的结构。结果 从南海海绵甲醇溶剂提取物中,分离鉴定了8个单体化合物:尿嘧啶(1)、胸腺嘧啶(2)、1 H-2-吡啶酮(3)、胸腺嘧啶-2’-脱氧核苷(4)、2-甲酰胺吡咯(5)、3-醛基吲哚(6)、6-羟基-4,4,7a-三甲基-5,6,7,7a-四氢苯并呋喃-2(2 H)酮(7)、5-氨基戊酸(8)。结论 化合物1~8均为首次从该种海绵中分离得到。化合物7在50μg·mL-1浓度水平,对K562白血病细胞株、P388白血病细胞株和HeLa肿瘤细胞株的抑制率分别是38.29%、23.23%和95.57%。
Objective To study the constituents of Agelas aff. nemoechinata. Methods The isolation and purification of the compounds were performed by Sephadex LH-20, silica gel, ODS chromatography and HPLC methods. And their structures were determined by comparison their physicochemical characters and spectral data with literatures. Results From the methanol extracts , eight compounds were isolated and identified as Uracil (1), Thymine (2), 1H-2-pyridone (3), 2-deoxythymidine (4), 2-pyrrole-carboxamide(5), 1H-indole-3-carbaldehyde(6), 6-hydroxy-4,4,7a-trimethyl-5,6,7,7a-tetrahydrobenzofu- ran-2(4H)-one(7) and 5-aminopentanoic acid (8). Conclusion Compounds 1-8 were isolated from this sponge for the first time. Compound 7 exhibited moderate cytotoxicity against K562, P388 and HeLa tumor cell lines with the proliferation inhibition rate 38. 29%, 23. 23% and 95.57%, at the concentra- tion level of 50 μg/mL, respectively.
