基于“组分结构”理论的8种墨旱莲乙酸乙酯提取物对NHBE细胞保护作用的比较 被引量:4

Comparison of protective effects of eight ethyl acetate extracts from Eclipta prostrate on NHBE cells based on component structure theory

摘要 基于"组分结构"理论对来源不同的墨旱莲乙酸乙酯提取物(EAEEP)中活性成分进行分析比较,研究其保护正常人支气管上皮(normal human bronchial epithelial,NHBE)细胞的物质基础构成。以MTT法比较不同EAEEP对香烟烟雾提取物(cigarette smoke extract,CSE)诱导的NHBE细胞保护作用,并采用UPLC分析不同EAEEP中酚酸、香豆草醚及黄酮类组分的含量。结果发现,8种EAEEP(0~200 mg·L-1)对NHBE细胞均有一定保护作用,并存在统计差异。其中,保护作用较强的是EAEEPⅦ和EAEEPⅧ,其EC50分别为46.97,56.29 mg·L-1。UPLC分析显示,8种EAEEP中酚酸、香豆草醚及黄酮类组分的含量存在显著差异。而作用最强的EAEEPⅦ(89.15 mg·L-1)和EAEEPⅧ(57.44 mg·L-1)质量总量却并非最高,最高的为EAEEPⅢ(132.25 mg·L-1)。结合"组分结构"理论,分析发现EAEEPⅦ和EAEEPⅧ中酚酸、香豆草醚及黄酮类组分的质量结构与其他样品显著不同,分别为1.0∶1.0∶0.5和1.0∶1.9∶0.8。提示EAEEP中可能存在特定的最优"组分结构"关系,可为其物质基础研究及质量控制提供参考。 To analyze and compare the protective effects of active components in different ethyl acetate extracts( EAEEPs)from Eclipta prostrate,in order to study the comparison of materials bases protecting normal human bronchial epithelial( NHBE) cells.The MTT assay was taken to compare the protective effect of different EAEEPs on cigarette smoke extracts( CSE)-induced NHBE cells.The ultra-performance liquid chromatography( UPLC) was applied to analyze the content of phenolic acid,coumaric grass ether and flavonoid in EAEEPs. According to the results,all of the eight EAEEPs( 0-200 mg·L- 1) showed certain protective effect on NHBE cells,with statistical difference. Specifically,the total mass of EAEEP Ⅶ( 89. 15 mg·L- 1) and EAEEP Ⅷ( 57.44 mg·L- 1),which showed the strongest activity,was not the highest,while EAEEP III( 132. 25 mg·L- 1) displayed the highest total mass. In the combination with the " component structure" theory,the analysis showed a significant difference in the mass structure among phenolic acid,coumaric grass ether and flavonoid in EAEEP Ⅶ and EAEEP Ⅷ,which were 1. 0∶ 1. 0∶ 0. 5 and 1. 0∶ 1. 9∶ 0. 8,respectively. The results suggested a specific optimal " component structure" relationship may exist in EAEEP,which could provide reference for the material base study and quality control.
