摘要
目的 在老年特发性肺间质纤维化(IPF)患者肺泡灌洗液游离囊泡(Exosome)中筛选出表达差异的微小RNA(miRNA).方法 收集10例老年IPF患者及10例健康志愿者的肺泡灌洗液并分离Exosome.进行miRNA分离,分离后获得的miRNA采用高通量miRNA表达差异定量检测阵列进行检测,检测结果采用GeneCopoiea在线工具进行分析.结果 高通量miRNA表达差异定量检测阵列显示,与健康对照组比较,在肺泡灌洗液的Exosome中miR-125b、miR-128、miR-21、miR-100、miR-140-3p、miR-374b与健康对照组比较,表达呈上调趋势,分别增高了5.42、9.93、7.62、5.86、7.80、4.16倍,let-7d、miR-30a、miR-27b和miR-103呈下调趋势,分别下降了2.41、2.87、3.27、3.45倍,差异有统计学意义(P<0.05).结论 老年IPF患者肺泡灌洗液Exosome中存在miRNA表达差异.
Objective In oder to select disease-related biomarkers,the differentially expressed microRNAs (miRNAs) from bronchoalveolar lavage fluid (BLAF) exosomes in elderly patients with idiopathic pulmonary fibrosis (IPF) patients were screened.Methods BLAF exosomes were collected from 10 cases of IPF and 10 healthy controls.The miRNA expression was checked by high-throughput quantitative detection of miRNA expression array,and the results were analyzed using online tools GeneCopoiea.Results miR-125b,miR-128,miR-21,miR-100,miR-140-3p and miR-374b in BALF exosomes from IPF patients were checked using high-throughput quantitative detection of miRNA expression array,and the expression of these miRNAs was upregulated respectively by 5.42,9.93,7.62,5.86,7.80 and 4.16 folds compared to the controls,and that of let-7d,miR-30a,miR-27b and miR-103 was downregulated respectively by 2.41,2.87,3.27 and 3.45 folds.Conclusion There is differential expression of miRNA in BLAF exosomes from IPF patients.
