8q24区域4个单核苷酸多态性与前列腺癌的相关研究 被引量:2

摘要 目的探讨染色体8q24区4个单核苷酸多态性(SNP)rs6983267、rs1447295、rs10090154和rs7837688与北京居民中前列腺癌(PCa)的关联及其基因型和表型的关系。方法收集138名正常对照者和124例PCa患者的血液样本和其遗传、嗜好、临床、膳食习惯等信息。基于高分辨率熔解曲线分析技术(HRM),对124名PCa患者和138名正常对照者的4个SNP(rs6983267、rs1447295、rs10090154和rs7837688)位点进行基因分型。结果 PCa与对照组间rs6983267(55.8%vs 56.8%)、rs1447295(21.5%vs 17.5%)、rs10090154(20.3%vs 16.4%)、rs7837688(19.7%vs 19.3%)各基因型分布差异无统计学意义(P>0.05)。数量性状分析发现,PCa患者中rs7837688的GG基因型携带者饮酒的酒精度数更高(P=0.022)。结论 8q24区rs6983267、rs1447295、rs10090154和rs7837688多态性与北京居民中PCa的易感性可能无相关关系。 Objective To explore the correlation between chromosome 8q24 (rs6983267,T), (rs1447295, A) , ( rs10090154, T), ( rs7837688, T) and prostate cancer (PCa) in Beijing residents, and to explore the correlation between genotype and phenotype in PCa patients. Methods 138 healthy controls and 124 PCa pa- tients phenotypes were collected including clinical , genetic , dietary habits, hobbies and blood samples. Based on high resolution melting analysis technique ( HRM ), genotype ( rs6983267, T ), ( rs1447295, A ), ( rs10090154, T) and ( rs7837688, T) between 124 PCa patients and 138 normal controls were carried out the cases and contrast analysis of the genes associated with prostate cancer. ResuLts rs6983267 ,T(case 55.8%, control 56.8% ), rs1447295 ,A( case 21.5% ,control 17.5% ) ,rs10090154 ,T( case 20.3% ,control 16.4% ) and rs7837688,T( case 19.7%, control 19.3% )of 8q24 were not significantly associated with PCa in the study sample( P 〉 0.05 ). Quantitative trait analysis showed that the alcohol degrees of rs7837688 ,T risk geno- types GG (47.67 ~ 7.34) in PCa patients was significantly higher ( P = 0. 022 ) than those in the other genotype carriers(26.75 + 19.67 ). Conclusion The genetic variation in 8q24 ( rs6983267, T), ( rs1447295, A), (rs10090154 ,T)and(rs7837688 ,T) is impossibly associated with PCa susceptibility.
