摘要
目的研究养营活血汤对肝纤维化大鼠的治疗作用以及对其血清TNF-α和IFN-γ水平的影响。方法选取60只SPF级的Wistar大鼠,随机分为6组:正常对照组,模型对照组,秋水仙碱治疗组,养营活血汤低、中、高剂量组。采用复合因素制备大鼠肝纤维化模型,12周后全部采用乙醚麻醉处死,心脏采血并在肝左叶同一部位取肝组织标本,HE染色,进行组织纤维化评定,并用ELISA法检测大鼠血清TNF-α和IFN-γ的含量,观察养营活血汤对其含量的影响。结果经养营活血汤低、中、高剂量治疗后大鼠肝组织纤维化增生均减少,汇管区内炎性细胞均减少,肝细胞水肿程度均明显减轻。肝纤维化分级显示秋水仙碱治疗组、养营活血汤中、高剂量组肝纤维化程度明显低于模型组(P<0.05或P<0.01);养营活血汤高剂量组肝纤维化程度明显低于养营活血汤低剂量组和秋水仙碱治疗组(P<0.01);养营活血汤中剂量组肝纤维化程度明显低于低剂量组(P<0.05)。细胞因子检测显示:养营活血汤中、高剂量组大鼠血清中IFN-γ的浓度与模型对照组相比,明显降低(P<0.01);养营活血汤低、中、高剂量组大鼠血清中TNF-α的浓度与模型对照组相比,明显降低(P<0.05或P<0.01)。结论养营活血汤可减轻大鼠肝纤维化程度,其作用与降低大鼠血清TNF-α和IFN-γ含量有关。
Objective To investigate the therapeutic function of construction - nourishing blood - activating decoction on hepatic fibrosis and its effect on serum TNF - α and INF -γ levels in rats. Methods A total of 60 specific pathogen free (SPF) Wistar rats were randomized into 6 groups as follows: normal control group, model control group, colchicine treatment group and construction -nourishing blood -activating decoction low dosage, medium dosage and high dosage treatment groups. Rat hepatic fibrosis model was established by com- pound factors. All rats were sacrificed by ether anesthesia 'after 12 weeks. Blood samples were taken by cardi- ac puncture. Liver samples were taken from the same place on the left lobe and the fibrosis grade was evalua- ted after HE staining. The effect of construction - nourishing blood - activating decoction on serum TNF -α and INF - γ were tested by ELISA. Results After the treatment with low, medium and high - dose of con- struetion -nourishing blood -activating decoction, a reduction was shown in hepatic fibrosis proliferation, in- flammatory cells in the portal area and severity of hepatic cell edema. Fibrosis staging showed significant lower fibrosis grade in colchicine treatment group, medium- dosage and high -dosage construction- nourishing blood - activating decoction groups, compared with that in the model control group (P 〈 0.01 ) ; fibrosis grade in medium - dosage group was significantly lower than that in the low - dosage group (P 〈 0.05 ). The cyto- kine analysis demonstrated decreased serum INF-γ concentration in medium and high -dosage construction - nourishing blood - activating decoction groups in comparison with that in the model control group (P 〈 0. 01 ) ; the serum TNF - α levels in all of the low, medium and high - dose construction - nourishing blood - activating decoction groups were lower than that in the model control group( P 〈 0.05 or P 〈 0. 01 ). Conclusion Construction - nourishing blood - activating decoction alleviates rat hepatic fibrosis grade, which is related with reduced serum TNF- α and INF-γ levels.
