消风宣窍汤对变应性鼻炎豚鼠模型IgE、IL-4、IFN-γ、组胺的调控作用

Regulatory Effect of Xiaofeng Xuanqiao Decoction on IgE, IL-4, IFN-γ, Histamine in Guinea Pig with Allergic Rhinitis

[目的]探讨消风宣窍汤治疗变应性鼻炎(Allergic Rhinitis,AR)的作用机理。[方法]将60只Hartley豚鼠随机分为正常组、模型组、消风宣窍汤高、中、低剂量组、氯雷他定组。卵蛋白(OVA)建立变应性鼻炎豚鼠模型,药物干预后,ELISA法检测各组豚鼠血清总IgE、IL-4、IFN-γ、组胺的水平。[结果]与正常组比较,模型组IgE、IL-4、组胺水平显著升高、IFN-γ水平明显下降(P<0.01);与模型组比较,消风宣窍汤高剂量组能显著降低血清总IgE、IL-4、组胺水平,升高IFN-γ水平(P<0.01),消风宣窍汤中、低剂量组、氯雷他定组能降低血清总IgE、IL-4、组胺水平,升高IFN-γ水平(P<0.05)。[结论]消风宣窍汤能促进IFN-γ的产生,抑制IL-4的形成,促进Th1/Th2分泌的细胞因子之间恢复平衡,从而减轻EOS的活化,减少IgE的合成,减少组胺的释放,从多方面阻止变应性鼻炎的炎症反应,达到治疗效果。

[Objective] Discuss the treatment mechanism of Xiaofeng Xuanqiao Decoction on guinea pig with allergic rhinitis.[Methods]Sixty guinea pigs were randomized into 6 groups： normal control group, allergic rhinitis（AR） group, high-dosage, equivalent-dosage, low-dosage Xiaofeng Xuanqiao decoction group, and loratadine group. AR guinea pig model was successfully established by ovalbumin（OVA） challenge method. Xiaofeng Xuanqiao decoction and loratadine were applied to intervene AR guinea pig. The serum levels of IgE and IL-4, IFN- γ, histamine were determined by ELISA methods. [Results]The expression levels of IgE and IL-4, histamine significantly increased,meanwhile the level of IFN- γ significantly lower compared with the normal group（P〈0.01）. Compared with that in AR model group, the serum level of IgE and IL-4, histamine in high-dosage Xiaofeng Xuanqiao decoction group significantly decreased,but the level of IFN-γ increased（P〈0.01）; the levels of IgE and IL-4, histamine decreased in other medication groups, meanwhile the level of IFN-γ increased compared with the model group（P〈0.05）.[Conclusion]Xiaofeng Xuanqiao can promote the produce of IFN-γ, inhibit the formation of IL-4, promote the secretion of Th1/Th2 cytokines to restore balance, to relieve eosinophil activation, reduce the synthet-ic of IgE, reduce the release of histamine, in many ways to prevent allergic rhinitis inflammation, to achieve therapeutic effect.
