两株野鸭源新城疫病毒分离株F基因和HN基因的分子特性分析 被引量:3

摘要 为了分析从湖南长沙分离到的野鸭源新城疫病毒(NDV)融合蛋白基因(F基因)和血凝素-神经氨酸酶基因(HN基因)分子特性,采用RT-PCR方法对分离自野鸭粪便的2株NDV的F基因和HN基因进行了扩增和遗传进化分析。F基因序列分析表明,编码区核苷酸为1 662bp,编码553个氨基酸,F基因碱性裂解位点序列为^112E-R-Q-E-R-L^117,符合NDV弱毒特征,与ClassⅠ类NDV代表毒株核苷酸同源性在92.3%-93.8%之间。HN基因序列分析表明,编码区核苷酸为1 851bp,编码616个氨基酸,与ClassⅠ类NDV代表毒株核苷酸同源性在91.6%-93.9%之间。根据F基因和HN基因遗传进化分析表明,此次分离到的2株NDV同属于ClassⅠ类Genotype 3型。 In order to analyze the molecular characteristics of F and HN genes of Newcastle disease virus(NDV),isolated from wild ducks in Changsha,Hunan province.The F gene and HN gene of two NDV,isolated from feces of wild ducks,were amplified by RT-PCR and genetically analyzed.The nucleotide length of F gene coding region is 1 662 bp,coding 553 amino acids.The motif of cleavage site of the fusion protein was ^112E-R-Q-E-R-L^117,which keeps consistent with the characteristcs of the reference Class I avirulent strains.Homology analysis of F gene revealed that sequence similarities with Class Ⅰ reference strains varied from 92.3%-93.8%.The HN gene contains a 1 851 bp open reading frame,coding 616 amino acids.Homology analysis of HN gene showed 91.6%-93.9%of identities between the two isolates and ClassⅠ reference strains.Phylogenetic analysis of the F gene and HN gene sequences revealed that the two isolates NDV are grouped into genotype 3in ClassⅠ.
