摘要
目的观察血管性痴呆(VD)大鼠小脑顶核形态学变化及活化型Caspase-3、PARP-1的表达。方法选用健康SD大鼠32只,随机分为假手术组(8只)及VD组(24只)。采用Morris水迷宫,在假手术组术后1周,VD组术后1、2、4周时对大鼠做行为学观察和测试,通过尼氏染色观察各组大鼠小脑顶核形态学的变化,用免疫组化法检测小脑顶核活化型Caspase-3、PARP-1的表达。结果 VD组大鼠各时间点水迷宫逃避潜伏期较假手术组均明显延长(P<0.01);尼氏染色可见VD组大鼠小脑顶核神经元细胞数较假手术组减少(P<0.01);VD组大鼠小脑顶核活化型Caspase-3、PARP-1的表达与假手术组比较明显增加(P<0.01)。结论 VD大鼠小脑顶核发生了损伤,活化型Caspase-3、PARP-1在此过程中发挥重要作用。
Objective To observe the morphological changes and expressions of cleaved Caspase-3 and PARP-1 in the cerebellar fastigial nucleus of rats with vascular dementia( VD). Methods Thirty-two healthy Sprague-Dawley( SD)rats were randomly divided into sham group( n = 8) and VD group( n = 24). Morris water maze test was performed to evaluate the behavior of rats in sham group at week 1 and VD group at three time points( week 1,2 and 4) after injury.The morphological changes of cerebellar fastigial nucleus were examined by Nissl staining. The expressions of cleaved Caspase-3 and PARP-1 in the cerebellar fastigial nucleus were detected by immunohistochemical method. Results Compared with sham group,water maze escape latencies of VD group at three time points increased significantly( all P〈0. 01). Compared with sham group,the number of neurons in the cerebellar fastigial nucleus significantly decreased in VD group( P〈0. 01). Compared with sham group,the expressions of cleaved Caspase-3 and PARP-1 in VD group significantly increased in the cerebellar fastigial nucleus( P〈0. 01). Conclusion The cerebellar fastigial nucleus is damaged in VD rats,while the cleaved Caspase-3 and PARP-1 play an important role in this process.
