黄芩素对肺炎支原体的抑制作用及对小鼠肺上皮细胞的保护机制研究 被引量:11

Study on inhibitory effect of baicalein on mycoplasma pneumonia and protection mechanism of pulmonary epithelial cells of mice

摘要 目的探讨黄芩素(Baicalein)对肺炎支原体(MP)抑制作用及其对机体保护机制,为MP感染的防治提供科学的实验依据。方法对经MP和Baicalein处理后的BALB/c小鼠肺组织进行HE染色,并进行病理组织学评分;采用Real-time Quantitative PCR测定Baicalein对MP的最小抑菌浓度(MIC);Real—time Quantitative PCR及WesternBlot分别检测BALB/c小鼠肺组织P,黏附分子mRNA及蛋白的表达情况;Reverse Transcription-PCR和免疫荧光法检测小鼠肺组织表皮生长因子(EGF)mRNA和蛋白的表达。结果 Baicalein能明显降低经MP处理过的小鼠肺组织病理学评分;Baicalein对MP的MIC为32μg/ml;Baicalein能明显下调P1黏附分子的基因转录和蛋白翻译、上调EGF基因转录和蛋白翻译。结论 Baicalein对MP有明显的抑制能力,并能通过抑制P1黏附分子的表达及促进EGF蛋白的表达来保护机体免受支原体的损伤。 Objective To investigate the inhibitory effect of baicalein on mycoplasma pneumonia and its the protective mechanism in body, and to provide scientific experimental basis for prevention and treatment of mycoplasma pneumoniae infection. Methods The mycoplasma pneumoniae and baicalein treated BALB/c mice lung tissues were stained with hematoxylin eosin ( HE), and histopathological grading. Minimum inhibitory concentration (MIC) of baicalein on myeoplasma pneumonia was determined by real-time quantitative polymerase chain reaction (PCR). The expression of P1 adhesion molecules mRNA and protein in lung tissue of BALB/e mice was determined with reverse transcription-PCR and Western blot. The expression of epidermal growth factor (EGF) mRNA and protein in lung tissue was detected by quantitative RT-PCR and immunofluorescence. Results Baicalein significantly reduced the mycoplasma treated mice′s lung tissue pathological score. The minimal inhibitory concentration of baicalein was 32 μg/ml. Baicalein significantly downregnlated Pl gene transcription and protein translation, and upregulated EGF gene transcription and protein expression. Conclusions Baicalein shows significant resistance to mycoplasma pneumoniae, and can protect the body against mycoplasma damage by inhibiting the expression of P1 protein and promoting the expression of EGF protein.
