摘要
采用均匀设计法优化了达托霉素发酵培养基配方,并对30L罐生产工艺进行了优化。确定达托霉素最优发酵培养基配方为:葡萄糖2.0%、淀粉2.0%、冷榨豆饼粉0.5%、蛋白胨0.6%;以该配方进行30L发酵罐生产,当前体癸酸钠的补料起始时间为发酵开始28h、补料总量为1.5%时,最终发酵效价提高了81%。对达托霉素的工业化生产具有重要的指导意义。
Uniform design was used to optimize the medium of daptomycin shaking flask fermentation.The process was also optimized in 30 L fermentor.The optimal fermentation medium was obtained as follows:glucose 2.0%,starch 2.0%,soybean flour 0.5%,peptone 0.6%.The fermentation process of daptomycin in 30 L fermentor with above optimum fermentation medium was optimized as follows:the initial feeding time of precurson sodium caprate was at 28 h,the total feeding amount was 1.5%.The final fermentation potency was improved by 81%,which was significant in industrial production of daptomycin.
