白花兜兰种子无菌萌发及试管成苗技术研究 被引量:13

摘要 本研究以白花兜兰种子为外植体,探讨不同添加物培养基对种子胚萌发的诱导、外源激素对继代增殖、生根培养的影响,并进行移栽试验。结果表明:有机添加物椰乳对原球茎的诱导有较好的促进作用;1/2MS+6-BA1.0+NAAA0.05对芽的诱导和增殖起明显的促进作用;1/2MS+IBA0.2 mg·L-1对白花兜兰的壮苗生根起关键性作用因素;碎树皮为栽培基质的移栽成活率达96%。 Using the seeds of Paphiopedilum emersonii as the explants, the effects of different organic additives on induction of protocorm, and that of plant growth regulators on the growth of subcultures were studied. The result showed that: 1 ) organic amendment of coconut milk significantly enhanced the induction of protocorm ; 2) the best culture medium for induction and proliferation of buds was 1/2MS + 6 - BA1.0 + NAAA0. 05 ; 3 ) the culture medi- um of 1/2MS + IBA0. 2 mg · L^-1 played the key role in the process of rooting and seedling growth; 4) transplants grown on the bark medium had a survival rate of up to 96%.
