摘要
本文以4-N-甲基溴蒽吡啶酮为原料,经一步取代反应,获得了一例共轭结构可用于活细胞核酸标记的荧光染料H-1。通过对染料分子H-1的基本性质表征和活细胞内核酸的荧光标记测定,验证了其对核酸的标记性能。实验结果表明:染料分子H-1是一例性能优异的荧光染料;同时该染料分子具有较低的生物毒性、光稳定性及水油两亲性,可用于活细胞内核酸标记。
Taking 4-- bromine-- N -- methylpyridone as raw materials, a conjugate structure fluorescent dye H--1 for the labeling nucleic acid in living cells were obtained after a one--step substitution reaction. The ability of fluorescent dye for the labeling nucleic acid was verified by basic properties characterization and the nucleic acid labeling in living cells experiment. The experiment results indicated that fluorescent dye H-1 have excellent performances. Furthermore, the fluorescent dye H-1 could be used to label nucleic acid in living cell with low cytotoxicity, good photosta- bility and appropriate water-oil amphiphilic.
