摘要 目的探索成年SD大鼠心室肌细胞急性酶分离及记录瞬时外向钾电流的方法。方法以成年SD大鼠为研究对象,利用Langendorff灌流系统,利用Ⅱ型胶原酶二步灌流方法分离细胞,利用膜片钳技术记录大鼠心室肌瞬时外向钾电流。结果镜下可见存活心肌:呈杆状或矩形,表面干净,横纹清晰,立体感强,能记录出瞬时外向钾电流达3000 pA。结论本实验所用的方法细胞成活率高,能记录出瞬时外向钾电流。 Objective To explore the method of acute isolate of separation of acute enzyme of ventricular myocyte and recording transient outward potassium current in adult SD rats. Methods Adult SD rats were selected as subjects, Lan-gendorff perfusion system and type II collagenase two step perfusion method were used to separate cell, and pacth clamp technique was used to record transient outward potassium current of ventricular myocyte in rats. Results The viable myocardium was seen Microscopically:it is lod or a rectangle,the surface is clean with clear cross striations and strong three-dimensional senseand it can record the transient outward potassium current up to 3000 pA. Conclusion Survival rate of cell of the method applied in the experiment and transient outward potassium current can be recorded.
