干扰睾丸Clock基因对小鼠体外受精的影响 被引量:1

摘要 目的:研究雄鼠睾丸Clock基因沉默后,小鼠体外受精的变化及机制。方法:通过在ICR小鼠睾丸内注射Clock干扰质粒(实验组)和HK阴性对照质粒(对照组),注射18天后收集雄鼠精子,计数精子浓度,前向运动精子百分率和精子总活率。再将孵育好的精子加入卵培养皿中,分别观察卵丘颗粒细胞驱散情况及卵裂情况,计算受精率、囊胚生成百分率和胚胎孵化率。结果:睾丸Clock基因沉默后,小鼠精子活动率及前向运动率较对照组降低(p<0.05),实验组精子驱散卵丘颗粒细胞能力的时间延长(p<0.001)。结论:睾丸clock基因可影响精子驱散卵丘颗粒细胞的能力,该机制在Clock基因影响小鼠生殖的过程中的作用有待进一步研究。
