摘要
目的研究大黄素对高脂血症金黄地鼠脂代谢的改善作用。方法将40只金黄地鼠随机分成4组,正常对照组(Con组)、模型对照组(HFD组)、阳性对照组(Simvastatin组,6 mg/kg)及大黄素组(emodin组,70 mg/kg),除Con组外,各组给予高脂饮食诱导形成高脂血症模型,并于给药治疗4周后测定血中三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)水平,收取血浆样品绘制脂蛋白谱及进行核磁共振代谢组学研究。结果 1高脂血症金黄地鼠给予大黄素治疗后,血中TG、TC和LDL-C含量显著降低,差异有统计学意义(P<0.05);2脂蛋白谱分析显示,大黄素可调节脂代谢紊乱金黄地鼠血清脂蛋白亚型的分布及不同脂蛋白亚型内的脂质含量,调节高密度脂蛋白脂质运载能力;3核磁共振代谢组学结果表明,大黄素可改善血浆内源性分子的代谢轮廓,其改善效果与辛伐他汀相当。结论大黄素具有改善高脂血症金黄地鼠的脂代谢紊乱的作用。
Objective To investigate the improvement effects of emodin on lipid metabolism in hyperlipidemia golden hamster. Methods 40 golden hamsters were randomly divided into four groups, namely, normal control group (Con), high fat diet group (HFD), Simvastatin (6 mg/kg) group, emodin (70 mg/kg) group. Except the control group, the other groups were induced hyperlipidemia by high fat diet. After 4 weeks of treatment, content of serum triglycerides (TG), total cholesterol (TC), low-density lipoprotein-cholesterol (LDL-C), and high-density lipoprotein-cholesterol (HDL-C), and plasma samples were drawn for lipoprotein and lipids profiles analysis and proton nuclear magnetic resonance spec-troscopy (1H-NMR)-based metabonomics analysis. Results ①Comparing with HFD fed hamster, emodin significantly reduced serum TC, TG and LDL-C levels (P〈 0.05). ②Lipid profile analysis showed emodin could regulate the pro-portion of lipoprotein subtypes and lipid content, suggesting the function of lipoprotein was improved by emodin. ③1H-NMR-based metabonomics study gave us highlight that, emodin and Simvastatin could improve the metabolic profile of plasma endogenous molecules to a similar degree. Conclusion Emodin can regulate lipid metabolism in hyperlipidemia hamster model.
