人参多糖对冷暴露卵巢细胞功能影响 被引量:3

Effect of Ginseng Polysugar on the Function of Luteal and Granulose Cells under Cold Exposure

摘要 目的探讨人参多糖对冷暴露大鼠卵巢功能的干预作用。方法采用体外细胞培养法分离培养大鼠卵巢黄体、颗粒细胞。设人参多糖组和实验组,在37、4、0℃环境温度下多次暴露,观察黄体与颗粒细胞分泌孕酮与环磷酸腺苷(cAMP)的变化,采用放免分析方法测定孕酮与cAMP生成含量。结果 4℃重复3次冷暴露,实验组黄体细胞与颗粒细胞孕酮和cAMP生成量呈升高趋势,cAMP升高明显(P<0.01);0℃第一次冷暴露,孕酮和cAMP生成量升高,cAMP升高明显(P<0.01),第2次和第3次冷暴露,均明显下降(P<0.05、P<0.01);人参多糖组增高和下降幅度明显减小,第2次和第3次0℃冷暴露后,孕酮和cAMP生成量明显高于实验组(P<0.05、P<0.01)。结论冷暴露强度、次数对大鼠卵巢黄体细胞与颗粒细胞分泌功能有影响,人参多糖对其具有适应性调节作用。 Objective To discuss the intervening effect of Ginseng Polysugar on the rat's ovary functions under the cold exposure. Methods To set the ginseng group and experiment group under the 37,4,0 ℃ cold exposure repeatedly,and through culturing the luteal and granulose cells of the rat's ovary in vitro. To study the variance of progesterone and cyclic AMP( cAMP) content which were excreted by luteal and granulose cells inducted by the human chorionic gonadotropin(hCG). To determine the progesterone and cAMP content is adopted through the radioimmunity. Results The progesterone and cAMP content excreted by luteal and granulose cells has a hoisting trend under the 4 ℃ cold exposure with three replications,and the cAMP content has a significant increase( P〈 0. 01). Under the 0 ℃ cold exposure,the progesterone and cAMP content has a hoisting trend in the first time and the cAMP content increases significantly( P〈0. 01),while in the second and third time,they both decrease significantly( P〈 0. 05,P〈 0. 01). The progesterone and cAMP content have a slighter variance in the ginseng group than in the experiment group,and increase significantly under both second and third 0℃ cold exposure( P〈 0. 05,P〈0. 01). Conclusion The progesterone and cAMP content excreted by luteal and granulose cells are influenced by the intension and times of cold exposure,which also are accommodated by ginseng polysugar.
