摘要
为探索调控山羊脂肪沉积的关键基因表达规律,筛选影响山羊育肥性状的候选基因,选取0月龄、6月龄和12月龄安徽白山羊,屠宰后取背最长肌、腿肌和胸肌检测肌内脂肪含量,用荧光定量RT-PCR分析磷酸酪氨酸互作结构域1基因(PID1)在安徽白山羊的心、肝、脾、肺、肾、小肠、脂肪、腿肌、背最长肌和胸肌等组织中mRNA的发育性变化。随着月龄的增加,肌内脂肪含量持续上升;同月龄背最长肌肌内脂肪含量显著高于腿肌和胸肌(P<0.05)。PID1基因在心、肝、脾、肺、肾、小肠、脂肪、腿肌、背最长肌和胸肌中均有不同程度的表达,12月龄脂肪组织中的表达水平大于6月龄脂肪组织中的表达水平,且PIDI基因表达水平与背最长肌肌内脂肪含量呈显著相关(r=0.968,P<0.05)。说明PID1基因与脂肪沉积性状显著相关,是影响肉羊育肥性状的候选基因。
To identify the expression rule of key gene controlling fat deposition and to screen the candidate gene af-fecting fattening in goat, Anhui white goats at 0, 6-and 12-month age were slaughtered to collect samples from longissimus dorsi muscle, crureus and pectorals for determining the intramuscular fat ( IMF) content. The developmental changes of phosphotyrosine interaction domain containing 1 ( PID1 ) gene mRNA expressed in heart, liver, spleen, lung, kidney, small intestine, muscles were detected by RT-PCR. The IMF content increased with the progress of development. The IMF content in longissimus dorsi muscle was significantly higher than those in crureus muscle and pectorals muscle ( P〈0. 05) at the same age. The PID1 expressions were ob-served in heart, liver, spleen, lung, kidney, small intes-tine and three muscles parts. The abundances of PID1 mRNA in adipose were higher in 12-month goat than those in 6-month goat ( P〈0. 05 ) , and were significantly corre-lated with IMF content in longissimus dorsi (r=0. 968,P〈0. 05 ) . It was indicated that PID1 was related to fat depo-sition and might be a candidate gene for goat fattening.
