摘要
人类恶性肿瘤的发生发展过程涉及多种基因的表达和功能异常。小干扰RNA(Small interfering RNA,siRNA)通过碱基配对的作用方式干扰特定胞浆内信使RNA(mRNA),从而介导基因沉默,其研究在肿瘤治疗的研究和应用中取得了突出进展。然而,siRNA体内给药仍存在诸多问题,相应的siRNA输送系统呈现出多样化发展趋势。本文总结归纳了肿瘤治疗研究中各种不同的siRNA纳米输送策略,包括提高肿瘤靶向性、增强细胞摄取效率和改善溶酶体逃逸能力等方面,旨在为设计新型siRNA纳米输送系统提供参考。
The onset and progression of human cancer involve the expression and functional abnormalities of a variety of genes. Small interfering RNA (siRNA) mediates gene silencing by way of interference effects of target messenger RNA with base pairing, which has made outstanding progress in research and application in cancer therapy. However, there are many problems for siRNA delivery in vivo. This review summarizes a variety of differ- ent siRNA delivery systems used for cancer treatment and some important strategies for improving siRNA transfection based on targeted accumulation in tumor, enhancing cellular uptake and improving endosomal/lysosomal escape. aiming to provide the reference for the design of siRNA delivery systems.
