摘要 研究酶液用量、pH值、温度、乙酸-1-萘酯用量等条件对酶促反应影响以及几种金属离子对酶促反应活性的影响,并建立酶抑制方程。结果表明:酶液用量0.25 mL、乙酸-1-萘酯0.6 mL、pH值6.5、温度38℃为酶促反应的最佳条件。Cu2+、Mn2+、Ca2+、Fe2+、Co2+等离子在一定浓度范围内对酶促反应活性具有激活作用,且Cu2+浓度为0.06 mol/L时激活作用最为明显,能提高60.37%。Co2+浓度为0.04 mol/L时次之,能提高51.75%。 The paper researched on the effects of enzyme dosage,pH value,temperature of reaction,aceticacid 1 -naphthyl acetate dosage,on enzymatic reaction ,studied the impact of several metal ions on the activity of the enzymatic reaction ,established equation of enzyme inhibition. The results showed that 0.25 mL amount of enzyme dosage, 0.6 mL amount of acetic acid 1-naphthyl acetate, pH value 6.5, temperature 38 ~C were optimum conditions of the enzymatic reaction. Cu2~.Mn2+,Ca2+,Fe2+.Co2+ and other inorganic metal ions have excitation effects on the catalytic activity of the enzymatic reaction within a certain range of concentration, and the effects 0.06 mol/L concentration of Cu2+ was the most obvious,it could increase 60.37%. And 0.04 mol/L concentration of Co2+ was second, it could increase 51.75%.
