摘要
目的筛选半边风抑制大鼠中性粒细胞呼吸爆发的有效部位,并探讨可能的机制。方法采用化学发光法测定乙醇提取物及各极性部位对大鼠中性粒细胞呼吸爆发的抑制活性;化学模拟机体内O-2·及H2O2的产生,采用化学发光法分别测定各样品对O-2·及H2O2清除作用。结果除石油醚部位(PEP)和萃余水部位(WAP)外,乙醇提取物(ACE)、乙酸乙酯部位(EAP)及正丁醇部位(BAP)对大鼠中性粒细胞呼吸爆发均具有不同程度的抑制作用,其IC50分别为41.4,19.7,80.3μg·ml-1;在活性氧清除实验中,仅EAP对O-2·和H2O2表现了较强的清除效果。结论半边风对大鼠中性粒细胞呼吸爆发具有较强的抑制作用,其活性部位为EAP和BAP,但二者的抑制机制可能不同。
Objective To observe the effects of ethanol total extract, PE - soluble, EtOAc - soluble, n - BuOH - soluble and watersoluble portions of Dendropanax chevalieri on rat neutrophil respiratory burst and explore the potentiai mechanism of anti - inflammatory of D. chevalieri. Methods By using phorbol myristate acetate (PMA) as the stimulant, luminol - dependent chemiluminescence was used to determine anti -PMN respiratory burst activity of different extracts of D. chevalieri. Similarly, their effects on free radicals generated by pyrogallol - luminol-carbonic acid buffer system and H2O2 - luminol - carbonic acid buffer system were observed.Results None but ethanol total extract, EtOAc - soluble portion and n - BuOH - soluble portion of D. chevalieri could inhibit rat neutrophil respiratory burst. And their IC50 are 41.4,19.7,80.3μg·ml^-1 respectively. However,only EtOAc - soluble portion show the activity in scavenging O2·and H202. Conclusion D. chevalieri indicated obvious anti -PMN respiratory burst activity.
