摘要
目的探讨葛根素对HepG2细胞胰岛素抵抗的改善作用。方法用MTT法检测葛根素对细胞增殖的影响;将HepG2细胞置于10μg/ml胰岛素培养液中24 h复制胰岛素抵抗模型,用葡萄糖氧化酶法检测葛根素对胰岛素抵抗HepG2细胞葡萄糖消耗的影响;采用蒽酮法检测细胞内糖原含量。结果葛根素对HepG2细胞增殖无显著影响,并可促进胰岛素抵抗HepG2细胞的葡萄糖消耗,增加肝细胞糖元含量。结论葛根素可调节HepG2肝细胞糖代谢,明显改善肝细胞胰岛素抵抗。
Objective To investigate the effect of Puerarin on insulin resistance in HepG2 cells. Methods M'IT was used to detect the effects of Puerarin on cell proliferation ; establishing the model of insulin resistance by placing the HepG2 cells in medium with 10μg/mL insulin for 24 h, studying the effect of Puerarin on the glucose consumption of HepG2 cells in insulin resistance by the method of glucose oxidase, and detecting intracellular glycogen content by the anthrone method. Results Puerarin had no effect on the proliferation of HepG2 cells, could promote glucose consumption of HepG2 cell in insulin resistance, and increased hepatic glycogen content. Conclusion Puerarin can regulate glucose metabolism of HepG2 ceils, and improved hepatic insulin resistance.
