红花夹竹桃寄生中芦丁和广寄生苷含量的高效液相色谱法测定 被引量:2

HPLC determination of rutin and guajeverin in Scurrula parasitica L. parasitizing on Nerium indicum Mill

摘要 目的建立HPLC法测定红花夹竹桃寄生中芦丁和广寄生苷含量的方法。方法采用HPLC法,以芦丁和广寄生苷为对照品,色谱柱为依利特Hypersil ODS柱(4.6mm×250mm,5μm),以乙腈-1%乙酸溶液梯度洗脱为流动相,流速为1.Om1·min^-1,检测波长365nm,测定红花夹竹桃寄生中芦丁和广寄生苷含量。结果芦丁、广寄生苷的线性范围均为8.24—329.6Iμg·ml^-1,相关系数分别为0.9995,0.9996,加样回收率(n=6)分别为101.76%,103.47%,RSD分别为1.34%、0.76%,红花夹竹桃寄生中芦丁和广寄生苷的平均含量分别为0.32%,0.11%。结论该方法准确、重现性好,可作为红花夹竹桃寄生药材的质量控制标准。 Objective To establish an HPLC method for the determination of the contents of rutin and guajeverin in Nerium indi-cure Mill. Methods Contents of rutin, guajeverin in Nerium indicum Mill was measured by HPLC a column of Hypersil ODS (4. 6mm×250mm,5μm),with mobile phase consisted of acetonitrile(B) -1% acetic acid(A) with the gradient elution,flow rate: 1.0ml· min^-1, detection wavelength of 365nm. Results The calibration curves showed good linearity in the range of 8.24 -329. 6 μg·mL^-1( rutin, guajeverin ) ; correlation coefficients were 0. 9995,0. 9996. The average recoveries ( n = 6 ) were 101.76 %, 103.47% ;RSD were 1.34% ,0.76% respectively. The average contents( n = 5 ) were 0.32% (rutin) ,0.11% ( guajeverin). Conclusion An accurate and reproducible method can be used for the quality control of Scurrula parasitica L. parasitizing on Nerium indicum Mill.
