三七种子无菌育苗技术的研究 被引量:1

Studies on the techniques of aseptic seedlings from Panax notoginseng seeds

摘要 目的研究获得生长整齐、健壮的三七幼苗的途径。方法将消毒后的三七种子置于培养基上进行无菌培养,对影响种子萌发和幼苗生长的因素进行优化。结果 20℃条件下培养60 d后,MS、1/2MS、B5、N6四种培养基中,MS培养基种子萌发率最高,达到80%,而N6培养基上种子萌发率最低;在种子培养过程中,暗培养条件下的种子萌发速度和萌发率均高于光照培养条件;去除果皮的种子出芽率最高,并且染菌率低。当将种子用赤霉素溶液处理,明显提高萌发率,20℃条件下培养45 d后,300 mg/L赤霉素溶液处理的种子出芽率接近90%。结论去掉三七种子的果皮,用浓度为300 mg/L的赤霉素浸泡后置于MS培养基中,在黑暗条件培养萌发效果最好。 Objective The experiments were conducted to obtain uniform and vigorous seedlings of San - qi. Methods The sterilized seeds of Panax notoginseng were cultured on artificial media, and then the factors affecting seed germination were investigated. Results Among the four kinds of examined media, the highest germination rate of seeds was achieved from the MS with a germination rate of 80% ,by contrast, the lowest was from the N6 ;the germination time and germination rate of seeds under dark were both better than that under illumination ; the seeds with removal of pericarp gained the highest germination rate and the lowest bacteria infection rate;when the seeds were treated with gibberellic acid, the germination rate would be improved, and could lead to a 90% germination rate after treated by 300 mg/L GA solution. Conclusion Remove Panax notoginseng seeds peel, after the seeds were treated with 300 mg/L GA solution, put it in MS medium,to develop the best effect in dark conditions.
