摘要
建立乳品中乳铁蛋白高效液相色谱的测定方法.采用pH值为7.0磷酸盐缓冲溶液做为提取剂,C18色谱柱分离,不同浓度的乙腈-0.5 mol/L的氯化钠溶液-三氟乙酸的二元系统进行梯度洗脱.每100 g奶粉(或每100 mL液态奶)中乳铁蛋白添加量为5~10 mg时,回收率大于90%.定量限为0.5 mg/100 g.该方法具有结果准确,操作简便,重复性好等优点,可用于乳品中乳铁蛋白的检测.
A method for the determination of lactoferrin in dairy products by high performance liquid chromatography (HPLC) was developed. Lactoferrin was extracted by the solution of pH 7.0 0.05 mol/L phosphate. The method was performed on C18 column, the analysis was performed by gradient elution using solvent systems acetonitrile-0.5mol/L sodium chloride-TFA. Lactoferrin content was in the range of 5-10 mg/100 g, the recovery was higher than 90%. The limit was 0.5 mg/100 g. The results indicated that this method is accurate, simple and reproducible, and thus suitable for the analysis of lactoferrin in milk products.
