毛细管电泳法同时测定三磷酸腺苷二钠注射液中苯甲醇与碳酸胍含量

摘要 目的:采用毛细管胶束电动色谱法同时测定三磷酸腺苷二钠注射液中辅料苯甲醇与碳酸胍的质量浓度。方法未涂层毛细管(50μm×67 cm,有效长度50 cm);压力进样,进样压力为3.4 kPa,持续时间为8 s,分离电压为25.0 kV,柱温为25℃;检测波长为200 nm;电极缓冲液为硼酸/十二烷基磺酸钠缓冲液( Borate/SDS Buffer,pH9.1),运行时间15 min。结果苯甲醇线性范围为0.0375-1.688 mg/mL( r=0.9969),回收率为104.7%-107.0%(RSD=0.9%,n=3),碳酸胍线性范围为0.04048-2.024 mg/mL(r=0.9994),回收率为95.7%-105.4%( RSD=3.2%,n=3)。结论本方法分离效果好、灵敏、可靠,可用于同时测定三磷酸腺苷二钠注射液中的2种辅料。 Objective To establish a micellar electrokinetic capillary chromatography ( MEKC ) method to determine benzyl alcohol and guanidine carbonate simultaneously in adenosine disodium triphosphate injection. Methods Samples were analyzed on a uncoated capillary of 50 μm ×67 cm(50 cm of effective length) , and injected hydrodynamically ( 8 s, 3. 4 kPa ) . The applied voltage was 25. 0 kV and the temperature was kept at 25 ℃. Borate/SDS buffer(pH9.1) was used. The detection wavelength was 200 nm,and the running time was 15 min. Results The linear range of benzyl alcohol and guanidine carbonate were 0.037 5-1.688 mg/mL(r=0.996 9) and 0.040 48-2.024 mg/mL(r=0.999 4),respectively. The recoveries of benzyl alcohol and guanidine carbonate were 104. 7%-107. 0%( RSD=0. 9%, n=3 ) and 95. 7%-105.4%(RSD=3.2%,n=3),respectively. Conclusion The proposed HPCE method,with the advantages of high efficiency,sensitivity and reliability,could be applied in simultaneous determination of two accessories in adenosine disodium triphosphate injection.
