摘要
目的应用MicroRNAs(miRNAs)芯片技术筛选鼻咽癌石蜡标本组织中差异表达的miRNAs,研究与鼻咽癌相关的miRNAs。方法从2例鼻咽癌石蜡切片和相配对的正常鼻组织中提取总RNA,miRNA芯片检测miRNA表达情况,然后在37例鼻咽癌石蜡切片和正常鼻组织中,用Real-time RT-PCR(Taqman)对部分差异表达miRNA进行验证。结果芯片结果显示:与配对的正常鼻组织相比,有24个miRNAs在鼻咽癌中异常表达,其中8个miRNA高表达,16个miRNA低表达。用Real-time RT-PCR(Taqman)验证显示,hsa-miR-205在鼻咽癌中表达水平升高(P=0.021);hsa-miR-34c在鼻咽癌组织中表达水平降低(P=0.019)。结论 hsa-miR-205和hsa-miR-34c可能参与了鼻咽癌的发生过程。
Objective To screen aberrant miRNAs in nasopharyngeal carcinoma by miRNAs microarray. Methods Total RNA was extracted from nasopharyngeal cancer tissues and adjacent noncancerous tissues from paraffin sections. The miRNA expression profiles were analyzed using a mammalian miRNA microarray. Aberrant miRNAs were then verified by real-time RT-PCR in 37 nasopharyngeal cancer and nontumorous tissues. Results 24 aberrant miRNAs were identified by miRNA microarray, including 8-high expression of miRNAs and 16-low expression of miRNAs. Among them, the expression of hsa-miR-205 was verified to be higher( P=0.021) and hsa-miR-34c was identified to be significantly lower in the cancer tissues compared to the normal tissues(P=0.019). Conclusion hsa-miR-205 and hsa-miR-34c may be involved in the process of nasopharyngeal carcinogenesis.
