TLR4对胃癌细胞放射敏感性的影响探讨

Radiosensitive Effect of TLR4 on Gastric Cancer Cells

目的探讨Toll样受体4(TLR4)对肿瘤细胞放射敏感性的影响。方法小鼠胃癌细胞株MFC分别经脂多糖(LPS)、瑞沙托维(TAK-242)和磷酸缓冲液(PBS)处理后进行5Gy照射和0Gy照射,分析MFC细胞TLR4表达情况、增殖活性、放射敏感性、凋亡率和细胞周期变化。结果 MFC细胞经5Gy X射线照射24h后TLR4表达水平显著下降;增殖活性、放射敏感性显著下降,凋亡率显著升高;细胞阻滞于G2/M期。经TAK-242和LPS处理后增殖活性、放射敏感性等均发生不同程度改变。结论 TLR4可对胃癌细胞放射敏感性产生较大影响。

Objective To explore the effect of TLR4 on radiosensitivity of gastric cancer cells.Methods The gastric cancer cell line MFC was irradiated with 5Gy and 0Gy X-ray after treated with LPS,resatorvid（TAK-242）and PBS,respectively.TLR4 expression,proliferation,radiosensitivity,apoptosis rate and cell cycle changes of MFC cells were analyzed.Results After 24 hours of 5Gy X-ray irradiation,the TLR4 expression level,proliferation,radiosensitivity of MFC cells were significantly reduced;apoptosis rate obviously increased and cells were blocked in G2/M phase.Proliferation activity and radiosensitivity changed in different degrees after TAK-242 and LPS treatment.Conclusion TLR4 has obvious radiosensitive effect on gastric cancer cells.
