摘要
目的了解重庆地区急性腹泻患儿博卡病毒(human bocavirus,HBoV)的基因型和流行特征,为HBoV相关腹泻的防控提供依据。方法以重庆医科大学附属儿童医院为哨点医院,收集2011年急性腹泻儿童患者的粪便样本504份,使用荧光定量PCR筛选阳性标本,使用巢氏PCR方法对阳性标本进行扩增,对扩增序列进行序列比对并建立进化树进行基因分型。结果 504份腹泻标本中,49份为HBoV阳性,阳性率为9.72%。不同季节HBoV的检出率不同,夏季检出率(19.72%)最高(P=0.003)。序列比对和进化树结果提示49份HBoV阳性标本中,HBoV1、HBoV2和HBoV3的例数分别25、23和1,其中检出的23份HBoV2全部为HBoV-2B型。结论不同季节HBoV的检出率不同,其中夏季病毒检出率最高;重庆地区检出的HBoV以HBoV1型和HBoV2B型为主。
Objective To investigate the prevalence, genotypes and epidemic characteristics of human bocavirus associated with acute pediatric diarrhea in Chongqing area. Methods Pediatric patients with acute diarrhea were recruited and stool samples were collected in Children' s Hospital of Chongqing Medical University during 2011. Real-time PCR and nested PCR were performed to determine the presence of HBoV and their genotypes. Results A total of 504 stool specimens were collected, from which 49 HBoV positive samples were identified, with a detection rate of 9. 72%. A seasonal distribution of HBoV was observed, with a peak ( 19.72% ) in summer. No distribution of age or gender specific was obtained for the HBoV positive detection. Based on Blast and MEGA analysis, 25 HBoV1, 23 HBoV2 and 1 HBoV3 were determined. All the HBoV2 strains detected belonged to HBoV-2B geno- type. Conclusions The detection rate of HBoV has obvious seasonality, with a peak in summer. HBoV1 and HBoV-2B are the major genotypes.
