摘要
目的探讨磷酸二酯酶抑制药多索茶碱对体外培养的急性髓系白血病(acute myeloid leukemia,AML)细胞U937增殖及凋亡的影响。方法分别将不同实验浓度的多索茶碱作用于一定浓度的处于对数生长期的U937细胞,采用WST-1方法检测细胞增殖抑制率,TUNEL法检测细胞凋亡。结果不同浓度的多索茶碱分别作用于U937细胞后,细胞增殖活性受抑程度随着作用时间的延长和药物浓度的增加而逐渐增加(P<0.01),其中以60 mg/L组最明显。不同浓度的多索茶碱均能促进U937细胞凋亡,且凋亡具有显著的浓度依赖性及时间依赖性(P<0.01)。结论多索茶碱能够抑制白血病细胞U937细胞株增殖,诱导其凋亡,并具有浓度及时间依赖性。
Objective To study the effects of the inhibitors of phosphodiesterase for acute myeloid leukemia in vitro culture U937 cell proliferation and apoptosis , and provide basis for the clinical adjuvant treatment of leukemia. Methods Add Doxofylline of different experimental concentrations was added respectively to a certain concentration of U937 ceils in ogarithmic phase. Cell prolifera- tion rate was detected by WST-1 method and the apoptosis by TUNEL method. Results Different concentrations of doxofylline acted on U937 ceils respectively. The degree of inhibition of cell proliferative activity gradually increased with prolonged action time and the increase of drug concentration( P 〈 0.01 ) , the most obvious among them was in 60 mg/L group. Different concentrations of doxofylline promoted U937 cell apoptosis and apoptosis with significant concentration dependence and time dependence( P 〈 0.01 ). Conclusions Confirmed by the experiment, doxo fylline can inhibit leukemia U937 cell line proliferation and induce its apoptosis, and has concen- tration dependence and time dependence.
