摘要
目的:研究人不同转移潜能前列腺癌细胞系PC-3M-2B4(低转移潜能)和PC-3M-IE8(高转移潜能)放射敏感性异质性及其机理。方法:通过流式细胞仪、荧光显微镜、电镜和Western-blot检测PC-3M-2B4和PC-3M-IE8凋亡能力的差别及凋亡相关蛋白Bcl-2、Bax、Caspase-3表达的差异,来证实人不同转移潜能前列腺癌细胞与放射敏感性的关系。结果:PC-3M-2B4和PC-3M-IE8两种细胞的凋亡率都随着照射后时间的延长而增加,但PC-3M-2B4比PC-3M-IE8凋亡率高,且差异有统计学意义(P<0.05);抗凋亡蛋白Bcl-2表达时间依赖性递减,而促凋亡蛋白Bax和Caspase-3表达明显升高。结论:人不同转移潜能前列腺癌细胞系,其转移能力与放射敏感性有关,转移能力低的对放射敏感性高。
Objective: To study the radiosensitivity heterogeneity of different metastatic potenhal cell hnes ot human prostate cancer PC-3M-2B4(low metastatic potential) and PC-3M-lE8(high metastatic potential) and Its mechanism.Method: To detect differences in PC-3M-2B4 and PC-3M-IE8 apoptotic ability, to detect the differences of apoptotie ability in PC-3M-2B4 and PC-3M-IE8 and the expression of apoptosis-related proteins Bcl-2, Bax and Caspase-3 by flow cytometry, fluorescence microscopy, electron microscopy and Western-blot.Result: The apoptosis rate of two cell PC-3M-2B4 and PC-3M-IE81 were irradiated with the extension of time increases.Compared to PC-3M-IE8, the apoptosis rate of PC-3M-2B4 was statistically significant(P〈0.05).The expression of auti-apoptotie protein Bcl-2 decline with time-dependent, while the expression of pro-apoptotic proteins Bax and Caspase-3 was significantly increased. Conclusion: Human prostate cancer cell lines with different metastatic potential, its ability of transfer has related to the and radiosensitivity, those who have low transfer ability with high radiosensitivity.
