摘要
对高地芽孢杆菌(Bacillus altitudinis)PY41所产碱性蛋白酶的酶学性质进行了研究.研究得出该蛋白酶的最适反应pH值为9.5,最适反应温度为50℃,米氏常数Km值为15.5 mg/mL,最大反应速度vmax为24.57μg/(min·mL).金属离子Ag+和Hg2+对该蛋白酶的抑制作用非常显著,使蛋白酶酶活降低了约90%,Mn2+对蛋白酶酶活具有明显的激活作用,使蛋白酶酶活力提高了18%.
This paper studied the enzymatic properties of alkaline protease produced by Bacillus altitudinis PY41.The results showed that the optimal pH of the protease was 9.5,the optimal temperature was 50 ℃,the Kmvalue was 15.5 mg/mL,and the maximum reaction speed vmaxwas 24.57 μg/(min·mL). Metal ion Ag+ and Hg2+ could inhibit the protease activity by about 90%;and metal ion Mn2+ could improve the protease activity by 18%.
