摘要
延龄草为国家三级保护植物,近几年,其资源数量急剧减少,急需寻找增加资源的途径.为了获得延龄草初代培养的无菌培养物,对延龄草初代培养中外植体所感染的微生物进行分离纯化、形态学及生理生化鉴定,进一步对污染外植体进行抑菌处理.结果表明,外植体所感染菌株均为埃希氏菌属(Escherichia);在培养基中加200 mg/L链霉素或300 mg/Lα-氨苄青霉素能获得较好的抑菌效果;100 mg/L链霉素或150 mg/Lα-氨苄青霉素浸泡污染外植体6 h也能获得较好的抑菌效果.该研究为延龄草离体繁殖体系的建立奠定了基础,从而为进一步从离体繁殖的角度扩大资源提供了可能.
Trillium tschonoskii Maxim is the national level 3 protection plants. In recent years, its present abundance contiunes to reduce. It is urgent to find a way to increase its resources. To obtain asepsis culture material in the initiation culture of Trillium tschonoskii Maxim, explant contaminated microorganism was isolated and purified, and further identified on the basis of morphology, physiology and biochemistry, and then the contaminated explants were treated with antibiotic solution. The results showed that the strain of bacterium of explant contaminated belonged to Escherichia. Better bacteriostasis was obtained by the supplementation of strep- tomycin at 200 or 300 mg/L, or ampicillin at 300 mg/L into the medium, or ahematively by soaking the contaminated explants in streptomycin at 100 mg/L or ampicillin at 150 mg/L for 6 hours. Our research paved the way to establish a vitro propagation system of Trillium tschonoskii Maxim. It is possible to further expand the resources in vitro propagation.
