不同体质类型冠心病心血瘀阻证患者的ApoE基因多态性分布 被引量:4

ApoE Gene Polymorphism Distribution in Coronary Heart Disease Patients with Blood Stasis Syndrome of Different Types of Constitution

摘要 目的:研究不同体质类型对冠心病心血瘀阻证患者ApoE基因多态性分布的影响。方法:运用王氏体质分类标准对600例冠心病心血瘀阻证患者进行体质分型,采用全序列基因测序法检测ApoE基因多态性,分析不同体质类型与ApoE基因多态性分布的关系。结果:冠心病心血瘀阻证患者血瘀质的主要ApoE基因型为E3/4(47.2%)、E3/3(33.2%)、E2/3(15.1%);气虚质的主要ApoE基因型为E3/3(50.0%)、E3/4(28.6%)、E2/3(11.9%);阳虚质的主要ApoE基因型为E3/4(51.7%)、E3/3(27.6%)、E2/3(15.5%)。各体质组的E3/3频率明显低于健康组,E3/4频率明显高于健康组。另外气虚质E3/3分布频率高于血瘀质和阳虚质,而血瘀质和阳虚质的E3/4高于气虚质。各体质等位基因频率均为E3>E4>E2,各体质组的E3明显低于健康组,而E4均高于健康组。各体质组之间等位基因比较,差异无统计学意义(P>0.05)。结论:E3/4、E3/3ApoE基因型可能为冠心病心血瘀阻证的易感基因型,在不同体质之间有一定差异,E3/4可能是血瘀质和阳虚质的易感基因型,而E3/3可能是气虚质的易感基因型;推测E3和E4可能是冠心病心血瘀阻证发病的遗传标记。 Objective: To study the ApoE gene polymorphism distribution in coronary heart disease patients with blood stasis syndrome of different types of constitution. Methods: The constitution of 600 patients with coronary heart disease patients of blood stasis syndrome were identified according to the "Wang"physique classification standard, and the ApoE gene polymorphism distribution of these patients were detected by Gene sequencing, to analyze the relationship between different types of constitution and ApoE gene polymorphism distribution. Results: The main ApoE genotype in coronary heart disease patients with blood stasis syndrome of blood stasis constitution were E3/4(47.2%), E3/3(33.2%) and E2/3(15.1%); E3/4(47.2%), E3/3(33.2%) and E2/3(15.1%) in Qi deficiency constitution; E3/4(51.7%), E3/3(27.6%) and E2/3(15.5%) in Yang deficiency constitution. The frequency of E3/3 in the constitution groups was lower than the healthy group and the frequency of E3/4 was higher than the healthy group significantly. The frequency of E3/3 in the Qi deficiency constitution was higher than that of the blood stasis and Yang deficiency constitution, while the frequency of E3/4 of blood stasis and Yang deficiency constitution was higher than that of the Qi deficiency constitution. The gene frequency sequence was E3 〉E4〉E2, and frequency of E3 was lower than the healthy group while frequency of E4 was lower than the healthy group, in the three types of constitutions. The comparison of allelic genes among the different constitution groups showed there was no statistical significance (P〉0.05). Conclusion: E3/4 and E3/3ApoE may be the susceptible genotypes of coronary heart disease patients with blood stasis syndrome, and showed difference in different types of constitution. E3/4 may be the susceptible genotypes of blood stasis and Yang deficiency constitution and E3/3 might be the susceptible genotypes of Qi deficiency constitution. It was surmised that E3 and E4 may be a genetic marker in pathogenesis of coronary heart disease with blood stasis syndrome.
