摘要
荷叶碱是荷叶中的一种阿朴啡型生物碱,为荷叶中主要的降脂活性成分。为能够充足的利用和提取荷叶中的荷叶碱,在酶辅助酸性乙醇提取荷叶生物碱的方法中,基于Box-Behnken响应曲面分析方法对酶解过程进行优化。研究酶添加量、酶解时间、酶解pH和酶解温度对荷叶生物碱提取效果的影响。结果表明,酶解最佳工艺条件为酶添加量0.14%、酶解时间4 h、酶解pH4.5和酶解温度45℃。在此条件下荷叶生物碱的提取率稳定,且与理论值接近,说明该响应曲面模型具有实际应用价值。
In the enzyme auxiliary acidic ethanol extraction to lotus leaf alkaloid method, response surface analysis method based on the Box-Behnken optimize the enzymolysis process. Study on the adding amount, enzymolysis time, pH and temperature on the effecting of lotus leaf alkaloid extraction. The results showed that the optimum conditions were enzyme 0.14%, enzymolysis time 4 h, enzymolysis pH 4.5 and enzyme solution temperature 45 ℃. Under the conditions the lotus leaf alkaloid extraction yield was stability, and closed to the theoretical value. All showed that the response surface model has practical application value.
