摘要
目的研究正加速度(+Gz)应激对不同程度冠状动脉(冠脉)狭窄小型猪模型血浆白细胞介素-6(IL-6)和心肌组织线粒体的影响。方法以巴马小型猪为研究对象,采用左前降支丝线环扎法建立不同程度冠脉狭窄模型,并使之暴露于高+Gz环境下,观察各组最大可耐受正加速度值以及离心前后IL-6水平的变化,同时取其左心室前壁心肌组织在电镜下观察不同组别心肌线粒体形态的变化。结果在最大可耐受+Gz暴露后各组模型猪IL-6水平呈线性升高;冠脉中度和重度狭窄模型猪IL-6的浓度较伪手术组均显著升高,轻度狭窄组则升高不明显。伪手术组线粒体形态正常;轻度狭窄组线粒体排列稍紊乱,其内嵴稍模糊;中度狭窄组嵴模糊、空泡化;重度狭窄组线粒体嵴明显模糊,空泡化较多,甚至破裂。结论最大耐受的+Gz对轻度冠脉狭窄的猪IL-6和心肌线粒体影响不大,中度和重度狭窄的猪IL-6升高显著,且心肌线粒体损伤明显;冠脉狭窄越重,IL-6升高越明显,线粒体损伤变化越重。
Objective To study the effect of interleukin-6(IL-6) and myocardial mitochondrial of miniature swine model accompanying coronary stenosis at different levels under positive acceleration(+Gz). Methods Bama miniature swine were taken as study object to establishing the model of left anterior descending wire cerclage artery stenosis at different levels,and exposed to +Gz,the maximum tolerated acceleration value and IL-6 levels before and after centrifugation were observed in each group; then myocardial mitochondria of anterior wall tissue of left ventricular were observed under electron microscopes. Results The IL-6 levels rise linearly each group exposed to the maximum tolerated acceleration. There is significant difference on IL-6 levels in different groups of porcine model exposed to the maximum tolerated acceleration,compared with the normal control group IL-6 levels in the moderate and the severe groups were significantly raised,but the mild was not. The mitochondrial morphology is nomal in contorl group. The mitochondrial arrange was disorderly slightly,the cristae was slightly fuzzy in mild stenosis group. The mitochondrial cristae was vague, vacuolation in moderate stenosis group. The mitochondrial cristae was significantly fuzzy,more vacuolation, and even rupture in severe stenosis group. Conclusions There was no significant change about IL-6 and myocardial mitochondria in mild stenosis group exposed to +Gz. There were IL-6 increased significantly and myocardial mitochondria injury obviously in the moderate stenosis group and severe groups; and coronary artery stenosis was heavier, IL-6 increased more significantly and myocardial mitochondria changed more obvious.
